in manual mode of lens it is working i mean asuarebox in viewfinder comes and i can take picture but when AF is enabled in lens it is not taking pictures

Have you tried with a different lens? Also, you can assign autofocus to another button, rather than the shutter button. have you done this by mistake?

Easy, you set your AF mode fromone shottoAI SERVO. Canon DSLRdo not show AF pointswhen it is in AI SERVO mode. Which is really annoying. I always enjoy seeing AF points light up on Nikon DSLR.
